FYI this is what the painting mode looks like now: Now we have our painting panel showing the mode’s tool palette: The only thing missing now is a control window to indicate where to paint. To create a new window we need to press Ctrl + P to bring up the ‘New Window’ dialog. Then we can type or paste the URL or file path to where we’re going to paint and select which mode to use: The ‘Create’ button will create a new window (the last one we created will close). And this is what our mode looks like now: And here is the ‘New Window’ dialog where we can create new windows. But that is not all, we need to make sure that the new window is maximized. Now let’s see the new window that we just created: We get the image to paint for our canvas. Now we need to zoom in, so that our paintbrush size is big enough. Here I’ve dragged the image to the left so it’s half of the way to the left. And the zooming button is the “Zoom” button at the bottom of the control window. The Art Brush size has been changed to 2.7, big enough to paint. Now we paint. Yep, that’s a lot of fun isn’t it? We have used in situ hybridization in the mouse to locate the insertions of two different retrotransposons, representing different members of the long terminal repeat (LTR) class of retrotransposons, in the imprinting control regions (ICRs) of chromosome 11. DNA from mice with different gene mutations that selectively delete different imprinted domains of this chromosome was hybridized to segments from the ICRs in order to analyze in detail the nature of the deletions. This analysis has allowed us to define the order of the sequences delimiting two different domains of silencing. These domains correspond to the target regions for the tightly localized (Igf2, H19) and the loosely localized (Snrpn) imprinted transcripts. In each case the imprinting control element (ICE) is inserted in a region that is methylated in the germ line, suggesting a direct role for DNA methylation in the imprinting process. The organization of the intact LTR sequences of the ICEs of the Igf2 and Snrpn domains has been analyzed. We show that there is striking similarity between both kinds of ICEs, and that the same ncRNA (ZAC RNA) is transcribed from these two different LTR sequences.
